Project: Molecular, metabolic and physiological responses to physical exercise in obesity treatment.

Project description: Currently, the coordinator has several approved projects and scientific collaborations aimed at investigating two mechanisms that determine individual responses to physical exercise. The studies integrate body composition, physical fitness, energy expenditure, metabolism, dietary intake, sound quality and circadian rhythms with analysis of DNA methylation, genetic expression, polymorphisms, metabolomics and exposome. Inflammation, oxidative stress, telomere compression, cortisol, bioactive lipids, mitochondrial metabolism, exercise and remodeling of muscle and adipose tissue have also been investigated. The integration of this information seeks to understand why each individual responds differently to exercise and contribute to more precise and personalized strategies for the prevention and treatment of obesity and other chronic diseases.

Project: Unpacking the role of the intestinal microbiota at the maternal-fetal interface: biomarkers and therapeutic agents for inflammatory and neurological conditions

Project description: The formation of the human intestinal microbiota begins during the intrauterine period and continues throughout the subsequent phases of extrauterine life. The microbiota plays a fundamental role in the predisposition and non-development of allergic, autoimmune diseases, as well as in various inflammatory processes. It is important to highlight that adequate colonization of the fetal intestinal system is influenced by the maternal microbiota, the type of delivery and the subsequent composition of the intestinal microbiota. In the subsequent phases of childhood life, maternal nutrition, diet and antibiotic use influence the state of eubiose versus dysbiose, which determines adequate growth and development from the neonatal period to adult life. Recently, there has been a lot of interest in the interaction between the intestinal microbiota, brain and pancreas, known as the intestine-brain and the intestine-pancreas. There are four proposed pathways through which intestinal bacteria communicate with the central nervous system (CNS): 1) a neural pathway mediated by the vagus nerve; 2) the immunological pathway, which consists of the modulation of the CNS immune system by the intestinal microbiota; 3) the endocrine pathway, through which hormones modulate intestinal permeability and microbiome composition; e 4) via two bacterial metabolites and neuroactive components. These neuroactive substances include neuropeptides, neurotransmitters and short chain fatty acids (SCFAs), all capable of modulating brain function. Probiotics are defined as live microorganisms that can provide benefits to the health of the host when administered in adequate quantities. Various pre-clinical and some clinical studies demonstrate evidence of two modulatory effects of probiotics on the brain, leading to the emergence of a new category called psychobiotics, but their exact action mechanisms are still largely unknown. This proposal, we intend to contribute to the understanding of these mechanisms, exploring the existence of a fifth mechanism with immunoregulatory effects, based on the effect of the maternal intestinal microbiota on the onset/deficiency of inflammatory and neurological diseases, such as endometriosis, type 1 diabetes (DM1), gestational diabetes and predisposition to depression in offspring. This new pathway could be mediated by extracellular vesicles (EV) or exosomes, which are released by bacterial or immunological cells, reaching intestinal sites and acting as immunoregulatory molecules.

Project: Associated taurine supplementation without multicomponent training: effect on white adipose tissue clearance and metabolic and molecular responses using a metabolomic approach in women with sarcopenic obesity

Project description: The development favors metabolic and body composition alterations that increase the risk of sarcopenic obesity. The project will validate the effects of taurine supplementation, associated or not with physical exercise, in women who are comfortable with this condition. The participants will be distributed into four groups: placebo, taurine, exercise associated with placebo and exercise associated with taurine. The intervention lasts 16 weeks. They will be evaluated by functional capacity, body composition, resting energy expenditure and food consumption. Blood tests and biopsy of the abdominal subcutaneous white adipose tissue will also be performed. We will investigate the clarification of the white adipose tissue, the metabolomic profile, the lipid metabolism, the inflammation, the oxidative stress and the methylation of DNA.
It is expected that the association between taurine and exercise will produce beneficial effects on the metabolism, muscle function and health of healthy women with sarcopenic obesity.

Project: Assessment of body composition, phase angle and biomarkers of oxidative damage to DNA in women with breast cancer.

Project description: Alterations in body composition, such as reduction of skeletal muscle mass and sarcopenic obesity, are associated with metabolic dysfunctions that can influence the clinical prognosis in women with breast cancer. In parallel, the oxidative profile presents implications on the integrity of DNA, evidenced by the accumulation of 8-hydroxy-2′-deoxyguanosine (8-OHdG) and on telomeric stability. This study aimed to longitudinally evaluate the association between body composition, phase angle and oxidative profile biomarkers, 8-OHdG and telomerase activity in women with breast cancer undergoing neoadjuvant treatment, comparatively to women without cancer matched by age and menopausal status.
